PROVENGE IMMUNOTHERAPY TREATMENT

Men diagnosed with advanced prostate cancer have a variety of life-extending treatment options to choose from. The choice of treatment depends on the stage of the cancer, as well as your age and overall health. It’s important for you to talk with your doctor to determine the best option for you.

Treatment Options May Include:

  • Hormone Therapy:  Blocks male hormones that prostate cancer cells need to grow.
  • Immunotherapy:  Activates a patient’s immune system to seek out and attack prostate cancer.
  • Chemotherapy:  Uses chemicals to stop prostate cancer cells from growing.

PROVENGE TREATMENT

PROVENGE is the only personalized treatment that is clinically proven to help extend life in certain men with advanced prostate cancer. It is an immunotherapy that takes your own immune cells and reprograms them to attack your advanced prostate cancer. This process delivers a treatment designed just for you.

In the clinical trial (IMPACT) that supported FDA approval, more men treated with PROVENGE lived longer than men not treated with PROVENGE.

  • More men in the PROVENGE group were alive 3 years after the start of the study than men not treated with PROVENGE
  • PROVENGE reduced the risk of death by 22.5% for patients in the PROVENGE group
  • Only 1.5% of men discontinued treatment with PROVENGE due to side effects

How is PROVENGE Different from Other Prostate Cancer Treatments?

  • PROVENGE is an immunotherapy—Immunotherapy works by stimulating the immune cells already in your body to attack prostate cancer cells.
  • PROVENGE is NOT chemotherapy—Chemotherapy uses chemicals to stop prostate cancer cells from growing.
  • PROVENGE is NOT a hormone therapy—Hormones are chemicals made by the body that control the activity of certain cells or organs. Hormone therapy in prostate cancer is used to decrease the level of testosterone in the body.

Patient Criteria for PROVENGE Treatment

PROVENGE therapy is for prostate cancer patients with metastatic prostate cancer, meaning that the cancer has spread beyond the prostate into lymph nodes, bone, etc. Additionally, eligible prostate cancer patients will have progression or growth of the prostate cancer despite the use of standard hormonal therapy.

Eligibility Guidelines for PROVENGE Therapy

  1. Castrate resistant prostate cancer (not responding to hormonal therapy)
  2. Bone scan or CT scan with evidence of prostate cancer spread to the lymph nodes or bones (but not lungs, liver, brain)
  3. Little or no cancer-related pain (no need for narcotic pain medications for cancer pain)
  4. Adequate organ function (liver, kidneys, bone marrow)
  5. Other considerations, including history of autoimmune disease or significant heart or lung disease

PROVENGE Treatment Process Overview

Each dose of PROVENGE is personalized by taking your own blood cells and reprogramming them to jumpstart your immune system. This section also talks about the importance of PSA levels and the role they play in the treatment process. Here’s a step-by-step overview of the process:

STEP 1: CELL COLLECTION

To make each dose, blood will be drawn from your body and passed through a machine that collects a small portion of your immune cells, along with some platelets and a small number of red blood cells. The machine returns the rest of the cells and blood to your body.

  • This cell collection process is called leukapheresis and takes 3-4 hours
  • Your cell collection will take place at a blood center with staff that have been trained to perform the procedure

STEP 2: PROVENGE TREATMENT

Your immune cells are then sent to a manufacturing facility to be made into a dose of PROVENGE specifically for you.

  • Each dose of PROVENGE is given about 3 days after your cell collection procedure, and your appointment will take about 2 hours.

After 6 appointments, your treatment with PROVENGE is complete. As the sample schedule shows, your time commitment for PROVENGE is only 6 appointments over about a month. No future treatments with PROVENGE are necessary after that.

The Importance of Keeping Scheduled Appointments

It’s important to keep all of your scheduled treatment appointments and arrive prepared. Your dose of PROVENGE is custom made for you and has a short shelf life. If you miss an infusion appointment, your personal dose of PROVENGE will expire and can no longer be used. As a result, you’d have to repeat the cell collection procedure so a new custom dose of PROVENGE could be manufactured.

We encourage you to use the calendar inside your Patient Comfort Kit to set reminders for yourself. You can also ask family members or friends to mark your procedure dates on their calendars too, to help ensure that you do not miss any appointments.

Information About PROVENGE and PSA Levels

The prostate-specific antigen (PSA) test is often used to monitor progression of prostate cancer and how effectively your treatment is controlling the disease. However in the PROVENGE clinical trial, the main goal was not to lower PSA levels—it was to extend life. So even if your PSA levels do not go down, PROVENGE may help you live longer.

Therefore you shouldn’t worry if your PSA levels don’t change, since changes in PSA levels are not indicative of how well your PROVENGE treatment is working. Be sure to talk to your doctor if you have any questions or concerns about your PSA levels.
